The interaction between EV71 2A and catalase

Popis: | 93 Infection with enterovirus 71 (EV71) can cause hand-foot-and-mouth disease, herpangina, and even encephalitis. EV71 is a member of Picornaviridae family. It has been demonstrated that EV71 2A protein can induce apoptosis in the cells, which should be related to viral pathogenesis. Yeast two-hybrid system was used to search for cellular protein(s) interacting with EV71 2A protein. The C-terminus of catalase protein was fished out by this approach. The binding domain of catalase with EV71 2A protein was also identified using the same system. Physical interaction between these two proteins was also demonstrated by GST pull-downed assay in vitro. Confocal microscopy also showed the same sub-cellular localization of these two proteins. N-acetyl cysteine (NAC) can block the cell apoptosis induced by EV71 2A. Thus, the cell apoptosis induced by EV71 2A should be through reactive oxygen species (ROS). The cell apoptosis induced by EV71 2A is major through protease dependent pathway, because EV71 2Amut induces less amount of cell apoptosis than wild type EV71 2A. Inhibition of catalase may increase oxidative stress and cause apoptosis in the cells. We propose that apoptosis induced by EV71 2A is through protease-dependent pathway(unidentified protein targets)and protease-independent pathway(i.e., catalase). |
