Preparation and characterization of chitosan nanoparticles from Curcuma extract

The emulsion-droplet coalescence method was used to prepare chitosan-Curcumins nanoparticles from the water and ethanol extracts of Curcuma powder. The effect of preparation conditions on the particle size and other properties of nanoparticles were investigated. When 0.1% chitosan and Curcumin extract were used with soybean oil as the oil phase, the minimum particle diameter was 231 nm. The maximum particle diameter was 8663 nm when 3.0 % chitosan and Curcumin extract were used with hexane as the oil phase. The result showed that nanoparticales prepared by using soybean oil as the oil phase was smaller than those prepared by using hexane as the oil phase. The largest amount of filtered solid particales, occurred as the Curcumins ethanol (95 ﹪) extract was used to prepare nanoparticales by using hexane as the oil phase.The other nanoparticales showed no apparent difference in filtatable solids. The Curcuma ethanol (95 ﹪) extract reduced all DPPH free radicals in test samples after 30 mins. Nanoparticles prepared by using hexane as the oil phase scavenged the free radical at a rate slower than those prepared by using soybean oil as the oil phase. The Curcumins standard solutions were unstable in buffers at pH 3.0、pH 7.2 and pH 9.0. The Curcumins ethanol (55 ﹪) extract was unstable in buffer at pH 9.0, but the Curcumins ethanol (95 ﹪) extract and the Curcuma deionized water extract were unstable in buffer at pH 3.0. The results showed that noparticales prepared by using soybean oil as the oil phase had a stability better than those prepared by using hexane as the oil phase. The color test result showed the yellow pigment loading efficiency of chitosan –Curcumins nanoparticles prepared by using soybean oil as the oil phase were higer than those prepared by using hexane as the oil phase. The Curcumins ethanol (95 ﹪) extract loading efficiency was smaller than the Curcumins ethanol (55 ﹪). This might be due to chitosan –Curcumins extract nanoparticles prepared by using 95 % ethanol had smaller particle diameter. In contrast, the chitosan –Curcumins nanoparticles prepared by using hexane as the oil phase the particle size was larger but the loading efficiency was lower.
