牛奶中粗脂肪之加速溶劑萃取測定與六種合成代謝性荷爾蒙之液相層析質譜分析研究

Two subjects including (1) extraction of raw lipid in powder milk by Accelerated Solvent Extraction (ASE) method and (2) simultaneous determination of six anabolic hormones by High Performance Liquid Chromatography/Atmospheric Pressure Chemical Ionization/Mass Spectrometry (HPLC/APCI/MS), were presented in the thesis. Drawbacks of traditional raw lipid quantitive methods are solvent waste, tedious and laborious. The aim of this study was to develop a green and efficient ASE approach. The results demonstrated that ASE is sufficient to replace conventional solvent extraction strategies because the recovery of ASE method is equivalent to the Chinese National Standard (CNS) approach. In the meantime, the approach just spent below 40 min of the whole procedure and 40 ml of the organic solvent in comparison with 8~10 hr and 400 ml approximately in traditional techniques. The innovative approach was constructed for determining carcinogenic endogenous (17β-estradiol, progesterone and testosterone) and synthetic (diethylstilbestrol, ethinylestradiol and zeranol) hormones by coupling solid phase extraction(SPE) with LC/APCI/MS. The recovery of one liter fortified real water sample at 1 μg/l level varied from 45~126% and the RSD varied from 2~11%. The limit of detection (LOD) spread in the range of 0.5 ng/l and 2.5 ng/l. The results described that waste water from boy or girl dormitories contained more testosterone and 17β-estradiol than other environmental water samples. The three synthetic hormones were absent in all of the samples collected in campus of National Tsing Hua University. In the study of human urine samples, the recovery of 10 ml fortified real sample at 10 μg/l level varied from 60~110% and the RSD varied from 0.8~11%. The limit of detection (LOD) distributed in the range of 0.2 μg/l and 0.7 μg/l. The results indicated that target analytes were absent in the samples of children below 10-year-old. Testosterone and 17β-estradiol were observed in the samples of adults and progesterone was detected in all of the women samples. The synthetic hormones were absent in almost all samples except an ethinylestradiol-containing sample of woman.
