On-line microwave hydrolysis and immunoaffinity electrospray mass spectrometry using sequential injection analysis system for morphine and morphine-3-glucuronide

Popis: | 90 An on–line microwave hydrolysis and immunoaffinity electrospray mass spectrometry using sequential injection analysis(SIA)was developed for the analysis of morphine and morphine-glucuronides in urine. The urine samples pretreated by off-line enzyme or acid hydrolysis to produce the morphine from its glucuronide derivates were here replaced by microwave assistant hydrolysis using SIA. The hydrolysis efficiency of morphine-3-glucuronide(M3G)in closed and opened microwave hydrolysis system is 92% for 6s and 95% for 80s respectively. The detection limit of morphine in urine was 100ppb(6s)and 200ppb(80s). The linearity from 100 to 4000ppb and 200 to 4000ppb were observed, respectively. The morphine-6-glucuronide can be efficiently hydrolyzed using this method. Furthermore, a system to analysis morphine and its glucuronides simultaneously was developed. The aimed was to determine morphine and its glucuronides derivates by immunoaffinity in SIA and analyzed by mass spectrometry. Thus, the hydrolysis pretreatment can be omitted. The detection limits are 50ppb for morphine and 300ppb for M3G and M6G, respectively. Linearity was observed in the concentration range 50 to 2000ppb(morphine)and 300 to 2000ppb(M3G and M6G)The higher throughput and automation can be achieved by these systems. The real urine samples from abused patients were studied and compared with the GC-MS method. |
