Cryopreservation of Bovine Oocytes

Experiments were conducted in an effort to develop a more effective open pulled straw (OPS) vitrification procedure for bovine oocytes. Different parameters likely to influence the survival of bovine oocytes after a vitrification procedure were evaluated:straw diameter, oocyte meiotic stage, exposed time in vitrification solution, sucrose concentration and combination of cryoprotectants. A reduction in the straw diameter from 1.7 mm to 0.4 mm increased the maturation rate from 37% to 54.3%. Oocytes were cryopreserved at different maturation stages(0, 6, 12 and 18 hr)showed a higher maturation rate at 6 hr and 12 hr(58.5% and 57.6%, respectively)than 0 hr and 18 hr(30.6% and 33.2%, respectively). To access EG-DMSO toxicity, the oocytes were equilibrated in EG and DMSO both at a concentration of 10% for 1, 5 and 10 min. The reduction maturation rates were showed(57.1%, 41.1% and 34.0%, respectively)when exposed time were increased. When bovine oocytes were exposed to different concentrations of cryoprotectants, the maturation rates were the highest with EG-DMSO group, than followed by EG-PG and PG-DMSO groups. In summary, oocytes were maturated in vitro for 6 hr, 0.4 mm straw diameter, 10% EG-10% DMSO for 1 min exposed time and 0.3 M sucrose with cryoprotectant resulted in an improved OPS system with 50% to 60% of oocytes maturation. Cleavage was achieved following insemination after oocytes vitrification and maturation. However, further experiments are needed to improve the cleavage rate and further pregnancy following embryo transfer.
