Study on the analysis of H2 histamine receptor antagonists by capillary electrophoresis.

A simple capillary electrophoretic method was established for the determination of cimetidine, ranitidine, famotidine and nizatidine in tablets. The background buffer was 100 mM sodium dihydrogen phosphate adjusted to pH 3.50. Analysis was run at the voltage of 20 kV and a detection wavelength of 214 nm. The method is sensitive with a detection limit of 10 mM (cimetidine), 20 mM (ranitidine), 20 mM (nizatidine) and 10 mM (famotidine) (S/N=3, hydrodynamic injection 1sec.). The calibration curves were linear over a concentration range of 100-300 mM. Mean recoveries were larger than 92%, while the intra-day and inter-day RSD and RE were all less than 5%. Head-column field-amplified sample stacking is an efficient, on-line sample concentration technique that can easily provide a sensitivity enhancement of three orders of magnitude. In this study, low concentration of H2-antagonists are effected across a water plug, with separation taking place in a binary buffer comprising 50% (v/v) ethylene glycol, 125 mM NaH2PO4 (pH 5.00), and detection is effected at 214 nm. Liquid-liquid extraction with ethyl acetate-isopropanol (96:4, v/v) is shown to provide extracts that are sufficiently clean. The calibration curves were linear over a concentration range of 0.1~1.5 mg/mL (cimetidine), 0.5~2.5 mg/mL (ranitidine), 0.5~3.0 mg/mL (nizatidine) and 0.1~0.9 mg/mL (famotidine). Mean recoveries were large than 91%, while the intra-day and inter-day RSD and RE were all less than 12%. The method is sensitive with a detection limit of 3 ng/mL (cimetidine), 30 ng/mL (ranitidine), 50 ng/mL (nizatidine) and 10 ng/mL (famotidine)(S/N=3, electric-driven injection 90 sec.). This developed CE method was applied to investigate the pharmacokinetic study of one volunteer dosing cimetidine tablet (200 mg).
