Effects of C60 fullerene derivatives on the free radical elicited damage in the hippocampus in vitro

The effects of C60 fullerneols 1 and sulfomatobutylate C60 derivated(C60.SO3Na), newer free radical trappers, in protecting against radical-induced injuries caused by hydrogen peroxide and cumene hydroperoxide were studied in hippocampal slices of rat in vitro. Both hydrogen peroxide(0.004-0.008%) and cumene hydroperoxide (0.005-1 mM) reversibly decreased theamplitudes of the CA1 evoked population spikes in the hippocampal slices at a concentration and perfusion time dependent manner. C60 fullerenols 1 and C60.SO3Na both significantly prevented the hydrogen peroxide (0.006%) and cumene hydroperoxide(1 mM) elicited decreasing effects of the population spikes. Adenosine (0.03 mM)and DNQX (0.002 mM) both reduced the amplitudes of population spikes. Neither C60 fullerenols 1 nor C60.SO3Na alter the decreasing effects of adenosineand DNQX. A thiobarbituric acid (TBA) test was used to asses the effect of C60 fullerenols 1 and C60.SO3Na on lipid peroxidation elicited in hippocampalslices by hydrogen peroxide and cumene hydroperoxide. Incubation in the presenceof C60 fullerenols or C60.SO3Na totally prevented the increase in lipidperoxidation by hydrogen peroxide and cumene hydroperoxide in both the incubationmedium and the slice homogenates. These results indicated that free radical trappers like C60 fullerenols 1 or C60.SO3Na prevented damage produced by hydrogen peroxide and cumene hydroperoxide in hippocampal slices. The effects may be associated with the free radical scavenging activities of C60 fullerenols1 and C60.SO3Na.
