Effect of Losartan on the Sexual Behaviors of Rats

It is an orally active drug to be used in the treatment of the hypertensive diseases. Many reports have been indicated that antihypertensive drugs may cause sexual dysfunction in patients. The present study carried out to understand the effect of losartan on sexual functions in rats. No changes of the intracavernous pressure ( ICP ) were observed in both spontaneously hypertensive rats ( SHRs ) and Wistar-Kyoto rats ( WKYs ) after acute or chronic treatment with losartan. These results suggest that the losartan did not affect on the erection in rats. In addition, the present study also found that basal tone of the isolated corpora cavernosa was not influenced by angiotensin II or losartan directly. The absence of angiotensin II receptors in the corpora cavernosa of rats can thus be considered. Then, the sexual behavior have been investigated in both SHRsand WKYs after chronic treatment with losartan ( 30 mg/kg/day, p.o. ). After the daily treatment with losartan, like the lowering of blood pressure, the mount latency was apparently increased in SHRs but not in WKYs. Nevertheless, other behavior parameters were not significantly changed in rats both SHRs and WKYs. Plasma catecholamine concentrations were measured by HPLC-ECD. Plasma concentrations of epinephrine was decreased by losartan for 1 month and 3 months in SHRs, while norepinephrine level was lowered in SHRs only after 3 months. It is suggested that the sympathetic tone was inhibited by losartan for chronic administration. The obtained results can clarify the influence of losartan in sexual functions which may help us to prevent the side effect of this compound before widely application in clinics. |
