Expression, Characterization and Application of Human T cell Lymphotropic Virus Type I Envelope Glycoprotein

Popis: | 81 The entire envelope gene of human T cell lymphotropic virus type I (HTLV-I) has been successfully expressed in insect cells using a non-fusion baculovirus vector. The recombinant envelope proteins were glycosylated and appeared to be proteolytically cleaved and had cleavage products corresponding in sizes to authentic envelope proteins. The recombinant proteins were shown to expressed on the surface of SF 21-AE insect cells by indirect membrane immunofluorescence assay. These surface molecules were biological active, as demonstrated by their ability to induce syncytium formation when cocultivated with HeLa cells. When probed with 36 HTLV-I-infected individuals' sera, the recombinant proteins gave all positive results, indicating the excellent antigenecity of the recombinant proteins. Our results indicated that the baculovirus expression system may provide a useful alternative approach to mammalian expression system for production of HTLV-I envelope protein. Also, since the baculovirus expression system produced several hundred times more envelope protein than HTLV-I infection, the high level of expression should allow the purification of large quantity of functional envelope proteins for subsequent immunological and biological examination of the ill- defined HTLV-I envelope glycoprotein and for development of new HTLV-I vaccine. |
