The Detection of 8-Hydroxy-2'-Deoxyguanosine (8-OHdG) in Artificial Urine

Popis: Commercially available, disposable screen-printed electrodes (SPEs) are a possible method of rapid and economic determination of the concentration of the biomarker 8-OHdG in artificial urine. The electrochemical response of 8-OHdG, a biomarker of ROS-induced stress, was investigated by both cyclic voltammetry and differential pulse voltammetry. The oxidation of 8-OHdG can be identified as quasireversible, and the linear response of the SPE for 8-OHdG was observed 0.001mM to 0.1mM in PBS, a sensitivity of 9.89 µA/mM, and the limit of detection was 0.0017mM. Variation between individual SPEs was found to be, on average, less than 8% for the range 0.001mM to 0.1mM. pH of solution heavily impacts the peak potential while the peak current remains unchanged (an inverse linear relationship between peak potential and pH was found). Multi-purpose artificial urine (MP-AU) was investigated using cyclic voltammetry and differential pulse voltammetry, resulting in the identification of one electrochemically active compound, uric acid. Uric acid has a similar oxidation potential to 8-OHdG, and with a greater concentration, the response of uric acid occludes that of 8-OHdG. Uricase was added to the MP-AU greatly reducing the concentration of uric acid, and DPV was used to quantify the concentration of 8-OHdG.
