Gluconeogenesis in the chicken has unique features due in part to the presence of two isozymic forms of PEPCK which have novel patterns of expression. Study of the molecular biology of the mitochondrial (PEPCK-M) and cytosolic (PEPCK-C) forms of PEPCK, and in particular, investigation of the extracellular stimuli responsible for altering their expression can enhance understanding of gluconeogenic flux in birds. Three overlapping cDNA clones for PEPCK-M from the chicken have been isolated and from these the primary amino acid sequence of PEPCK-M has been deduced. Three potential AUG codons have been identified, as well as several unique features within the 3′ untranslated region of the PEPCK-M cDNA. Comparisons have been made between the nucleotide and amino acid sequences of PEPCK-M and the corresponding PEPCK-C sequences in the chicken. Northern analysis of mRNA extracted from livers of chick embryos and adult chickens has revealed the presence of two discreet mRNAs for PEPCK-M, 4.2kb and 3.4kb in size. |
