INDICATORS OF THE PROTEINASE SYSTEM/PROTEINASE INHIBITORS IN ACUTE EXPERIMENTAL PERIODONTITIS AND AFTER CORRECTION BY STEM CELLS

Popis: The purpose of the study was to investigate the effect of stem cells on the activity of proteolysis processes in rats with periodontitis, corrected by stem cells. The results of the study of the proteinase system/proteinase inhibitors in animals with acute experimental periodontitis, which was corrected with the help of human mesenchymal stem cells, were analyzed. Depending on the treatment, the animals were divided into several groups: 1st group – animals without pathology (intact); 2nd – animals with simulated acute periodontitis; 3rd – animals with periodontitis corrected by mesenchymal stem cells. The observation dates were set on the 1st, 7th, 14th, and 21st days after the animals were removed from the experiment. Indicators characterizing the activity of proteolysis processes were determined (by lysis of azoalbumin, azocasein, and azocol, using a set of “SimkoLtd” (Ukraine) reagents and proteolysis inhibitors (α2-MG, α1-IP), and the proteolysis index was also calculated. According to the results of the study, it was established that during the experimental modeling of acute periodontitis, there was a pronounced activation of proteolysis with a maximum on the 1st day of observation and a slight increase in the level of protease inhibitors. A less pronounced increase in inhibitory potential, along with a significant increase in proteolytic activity, was reflected in a significant increase in the value of the proteolysis index. The use of MSCs reduces the imbalance of the protease/antiprotease system, which is accompanied by a decrease in the proteolysis index, especially in the later periods after injury.
