Novel method for the synthesis of Sulfonamide Urea’s from p-toluene sulfonyl isocyanate using Amberlite IRA-400 Cl resin: Application towards the synthesis of Gliclazide

Popis: Novel and facile method for the synthesis of sulfonyl urea’s from p-toluene sulfonyl isocyanates with different amines using Amberlite IRA-400 Cl resin in NMP was reported. Different aromatic, aliphatic and heterocyclic amines were used to test the reaction conditions and most of the amines were well tolerated to form their corresponding sulfonyl urea’s in good to better yields. Using this methodology Gliclazide, an anti-diabetic drug was synthesized in 88% yield. Simple operational procedure, less reaction time, no chromatographic purification makes this methodology simple and practical.
