To observe the effect of astaxanthin (ASTA) on the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) in suspended leukocyte-depleted red blood cells stored for transfusion.The suspended leukocyte-depleted red blood cells were randomly divided into group A, B, C and D. The ASTA was added into preservation solution of suspended leukocyte-depleted red blood cells of group B, C and D with the final concentration 5, 10 and 20 μmol/L, respectively, while DMSO was added into cells of group A in the same volume. After 7, 14, 28 and 42 days of storage, the reactive oxygen species (ROS) content in red blood cells was detected by fluorescence microplate reader, malondialdehyde (MDA) content was detected by thiobarbituric acid (TBA) method, activity of SOD was detected by xanthine oxidase method, the activity of CAT was detected by visible light method, and activity of GSH-Px was detected by colorimetry.After 7, 14, 28 and 42 days of storage, the contents of ROS and MDA in suspended red blood cells of group B, C and D were significantly lower(P0.05), while the activities of SOD and GSH-Px were higher than those of group A(P0.05); and CAT activity in cells treated by ASTA was significantly higher at 28 and 42 days of storage in comparison with that of group A(P0.05). There were positive correlations between the ROS, MDA content in suspended red blood cells of group A, B, C, D and storage time(P0.01), while negative correlation between SOD, CAT, GSH-Px activity and storage time(P0.01).ASTA can decrease the oxidative stress level and peroxide damage degree by increasing the antioxidant enzyme activities in suspended leukocyte-depleted red blood cells during storage.虾青素对储存去白悬浮红细胞内抗氧化酶活性的影响.观察抗氧化剂虾青素(ASTA)对储存去白悬浮红细胞内超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)等抗氧化酶系活性的影响.去白悬浮红细胞随机分为A、B、C、D 4组, B、C、D组悬浮红细胞保存液内加入ASTA,其终浓度分别为5、10和20 μmol/L,A组只加入ASTA的溶解液DMSO。去白悬浮红细胞储存至d 7、14、28和42时,荧光酶标仪测定红细胞内活性氧族(ROS)含量,硫代巴比妥酸比色法测定细胞内丙二醛(MDA)含量,黄嘌呤氧化酶法测定SOD活性,可见光法测定CAT活性,比色法测定GSH-Px活性.储存至d 7、14、28和42时,B、C、D组悬浮红细胞内ROS和MDA含量均明显低于A组(P<0.05),SOD活性、GSH-Px活性均明显高于A组(P<0.05);储存至d 28和42时,CAT活性均明显高于A组(P<0.05)。A、B、C、D 4组去白悬浮红细胞内ROS含量、MDA含量与储存时间均呈正相关关系(P<0.01),SOD活性、CAT活性、GSH-Px活性与储存时间均呈负相关关系(P<0.01).ASTA通过提高去白悬浮红细胞内抗氧化酶系活性来降低细胞内氧化应激水平和过氧化损伤程度. |
