POLYMERASE GENE MUTATION PATTERN IN PATIENTS OF CHRONIC HEPATITIS B PRETREATMENT RSU WAHIDIN SUDIROHUSODO

Popis: Hepatitis B is caused by infection with hepatitis B virus (HBV) is a health problem in the world including in Indonesia, the emergence of mutations in the polymerase gene (P gene) HBV can lead to viral resistance to antiviral drugs such as analog nukleos(t)ide. This study aimed to determine the pattern of mutations in the P gene on Pre-treatment of chronic hepatitis B patients. HBV genomes isolated from 72 patients Sudirohusodo Wahidin Hospital, Makassar. HBV DNA is used for detection, amplification and sequencing P gene. P gene amplification was successfully performed on 23 patients (45%), and 19 (82.6%) of them had a mutation in the P gene. Found 1 mutation pattern in 1 patient that can lead to viral resistance to antiviral namely V207M pattern. In addition the most pattern of amino acid substitutions found in P genes is rtL269I P (52.63%), rtT332N (52.63%), rtQ238H (47.36%), and rtM145L (42.10%). Concluded that the pattern of mutations causing resistance to antiviral can be found in 1 patient Pre-treatment while the other patient who are not found show that the treatment can be started.
