Induction of the mesenchymal stem cell to the skeletal muscle cell by the effect of growth factors

Popis: OBJECTIVES: Theobjektive of this study was to evaluated the differantitaion of humanadipose tissue derived mesenchymal stem cells (MSCs) into skeletal muscle cellsby the effect of IGF1 and FGF2 growth factors. METHODS: MSCswere divided into four groups. I.group (control group) was incubated only inmyogenic medium, II. group was incubatedby adding 100 ng/ml FGF2 to the myogenic medium, III. group was incubated byadding 20 ng/ml IGF1 to the myogenic medium, IV. group was incubated by adding100 ng/ml FGF2 and 20 ng/ml IGF1 to the myogenic medium. Cells dyed with MyoD1antibody were analyzed by flow cytometry so as to determine myogenicdifferentiation at the end of 28 days.RESULTS: Itwas confirmed that 11.5% of the control group, 39.2% of the FGF2 group, 23.1%of the IGF1 group and 39.3% of the IGF1-FGF2 group showed differentiation. CONCLUSION: Ourresults show that IGF1 and FGF2 have a positive contribution on myogenicdifferentiation of MSCs, contribute to the studies related to muscle diseasesand treatment of these diseases to support by growth factors the feature of theregeneration capacity and differentiation.
