Popis: | Iako je klinička praksa pokazala kako opsežne ozljede oralne sluznice često zaraštavaju bez ožiljka, čak i kada zahvaćaju bazalnu laminu, razlog ove pojave nije dugo bio objašnjen. Jedan od osnovnih uzroka brze obnove bi mogla biti nedavno otkrivena populacija visoko potentnih matičnih stanica sluznice usne šupljine. Ove multipotentne stanice pokazale su veliki potencijal diferencijacije u brojna tkiva, što predstavlja značajnu osnovu za potencijalnu terapijsku primjenu. Njihovo podrijetlo iz neuralnog grebena čini ih etički vrlo prihvatljivim izvorom za matične stanice koje se mogu diferencirati u neurone. Cilj ovoga istraživanja bio je odrediti utjecaj hipoksije na matične stanice oralne sluznice. Pokazali smo kako stanice izražavaju biljege matičnosti, a izlaganje ovih stanica hipoksiji rezultiralo je smanjenom aktivnosti biljega OCT4 i Sox2. U atmosferskom kisiku nestin je bio distribuiran u citoplazmi i jezgri stanice, no prilikom hipoksije nestin pokazuje tedenciju redistribucije u jezgru. Although clinical practice has shown that extensive oral mucosal injury often heals without scarring, even when the basal lamina is affected, the cause of this phenomenon has not been well documented. The main cause of this rapid recovery is today considered to be the existence of a recently discovered population of highly potent stem cells of the oral cavity. These multipotent cells have shown a huge potential for differentiation into numerous tissues, offering significant therapeutic potential. Their neural crest origin makes them an ethically acceptable source for stem cells that can be differentiated into neurons. The aim of this study was to determine the effect of hypoxia on stem cells of the oral mucosa. We have shown that cell express stemness markers, however, exposure of these cells to hypoxia resulted in reduced expression of OCT4 and Sox2 markers. In the atmospheric oxygen nestin was distributed in the cytoplasm and the nucleus of the cell, yet in hypoxia nestin shows tendency to redistribute in the nucleus. |
