The effect of antilipid drugs on butyrylcholinesterase in biological samples of rats

Popis: | Butirilkolinesteraza (BuChE) se sintetizira u jetri i odmah izlučuje u krv. Osim u plazmi i jetri, prisutnost enzima je dokazana i u mozgu. U središnjem živčanom sustavu BuChE hidrolizira acetilkolin i poput acetilkolinesteraze važna je za normalnu kolinergičku funkciju. Oba enzima se u mozgu povezuju s kolinergičkim deficitom i progresijom demencije. Stoga se lijekovi koji inhibiraju katalitičku aktivnost kolinesteraza koriste za liječenje Alzheimerove bolesti. Učinci antilipidnih lijekova na aktivnost BuChE plazme i mozga su suprotni, tj. mogu stimulirati ili inhibirati njezinu aktivnost. Stoga su ciljevi ovog rada bili u Wistar štakora utvrditi učinak višekratne primjene različitih doza rosuvastatina, simvastatina i fenofibrata na aktivnost BuChE mozga, plazme i jetre. Male doze oba statina i obje doze fenofibrata nisu utjecali na aktivnost BuChE mozga, dok su je veće doze oba statina malo ali značajno povećale. Sva tri lijeka izazvali su značajan porast aktivnosti BuChE u plazmi, dok je u jetri značajan porast aktivnosti izazvao samo fenofibrat. Prema ovim rezultatima smatramo da je utjecaj antilipidnih lijekova na aktivnost BuChE štetan, što je od značaja za bolesti središnjeg živčanog sustava s oštećenom kolinergičkom funkcijom. Butyrylcholinesterase (BuChE) is synthesized in liver and released into plasma immediately after synthesis. BuChE is also presented in brain where hydrolyses acetylcholine and, like acetylcholinesterase, is important for normal function of cholinergic system. In brain both enzymes are associated with cholinergic deficit and progression of dementia. Therefore drugs that inhibit enzyme activity are used for treatment of Alzheimer's disease. The aims of study are to etsablish the influence of multiple applications of rosuvastatin, simvastatin and fenofibrate on BuChE activity in brain, plasma and liver of Wistar rats. In brain, small doses of both statins and fenofibrate in both doses haven't changed the BuChE activity, but high doses of both statins have caused a little but significant increase of enzyme activity. Increase of plasma BuChE activity was significant after administration of all three drugs. Fenofibrate has caused the singnificant increase of BuChE activity in liver. According to these results we suggest that the increase of BuChE activity is the side effect of anilipid drugs and is important for the illnesses of central nervous system with disturbed cholinergic function. |
