Imunohistokemijska lokalizacija ekspresije faktora rasta VEGF, FGFR1 i FGFR2 tijekom razvoja fetalnog bubrega čovjeka

Popis: | Osnovni cilj ove studije je imunohistokemijski ispitati ekspresiju vaskularnog endotelnog čimbenika rasta (VEGF) i receptora fibroblastnog čimbenika rasta 1 i 2 (FGFR1 i 2), u fetalnim bubrezima kako bi se utvrdila prostorna (glomeruli, proksimalni i distalni kanalići) i vremenska (od 18. do 37. tjedna razvoja) distribucija ekspresije navedenih čimbenika kao potencijalnih determinanti razvoja bubrega. Tkivo fetalnog bubrega uklopljeno u parafin i analizirano metodom imunofluorescencije pomoću antitijela na VEGF, FGFR1 i FGFR2. Podaci su analizirani različitim testovima: „Kruskal-Wallis“, „Dunn's Multiple Comparison“, „Tamhane's T2 multiple comparisons“, „Brown-Forsythe ANOVA“, „Welch's ANOVA“ testom i testom za linearni trend. VEGF, FGFR1 i FGFR2 su eksprimirani tijekom svih istraživanih tjedana razvoja fetalnog bubrega što potvrđuje njihov utjecaj i u ranijim i u kasnijim stadijima. Također je dokazana njihova kolokalizacija (prekrajanje). Uočeno je smanjenje ekspresije svih čimbenika, u svim tkivima, od 18. prema 37. tjednu razvoja i testovima je dokazana prisutnost statistički značajne razlike među pojedinim tjednima i pojedinim tkivima. Prisutnost VEGF, FGFR1 i FGFR2 tijekom razvoja humanog fetalnog bubrega dokazuje njihovu potencijalno bitnu ulogu u formiranju organa i funkciji istog. U budućnosti bi se trebalo više posvetiti istraživanju uloge ovih molekula u glomerulima, proksimalnim i distalnim zavijenim kanalićima. The aim of this study is to immunohistochemically examine the expression of vascular endothelial growth factor (VEGF), and the expression of fibroblast growth factor receptors 1 and 2 (FGFR1 and 2), in fetal kidneys to determine spatial (glomeruli, proximal and distal ducts) and temporal (from 18th to 37th week of development) expression of these factors as potential determinants of kidney development. Fetal kidney tissue embedded in paraffin was stained with VEGF, FGFR1 and FGFR2 antibodies by double immunofluorescence. Data were analyzed by various tests: Kruskal-Wallis, Dunn's Multiple Comparison, Tamhane's T2 multiple comparisons, Brown-Forsythe ANOVA, Welch's ANOVA test and the Linear Trend Test. VEGF, FGFR1 and FGFR2 were expressed during all of the studied weeks of fetal kidney development, confirming their impact at both earlier and later stages. Their co-localization has also been proven. A decrease in the expression of all factors, in all tissues, over time was observed and the presence of a statistically significant difference between weeks and tissues was demonstrated by tests. The presence of VEGF, FGFR1 and FGFR2 during the development of human fetal kidney proves their potentially important role in organ formation and function. In the future, more attention should be devoted to investigating the role of these factors in glomeruli, proximal and distal convoluted ducts. |
