Impact of obesity and lifestyle factors on gut microbiota

Popis: Pretilost, koja je danas sve češća pojava, se povezuje s razvojem kroničnih bolesti i povećanim mortalitetom. Zbog prevencije pretilosti važno je otkriti čimbenike koji utječu na njen razvoj. Pretilost se počela povezivati s promjenom u sastavu crijevne mikrobiote. Cilj rada bio je provjeriti povezanost indeksa tjelesne mase (ITM) i životnog stila sa sastavom crijevne mikrobiote. U rad je uključeno 25 pretilih (ITM > 27 kg mˉ²) i 15 pothranjenih (ITM < 20 kg mˉ²) ispitanika koji su ispunjavali opći upitnik o životnom stilu. Sastav crijevne mikrobiote je određen izolacijom DNA iz uzoraka fecesa ispitanika i njenim sekvencioniranjem na Illumina MiSeq uređaju. Sirovi podaci sekvencioniranja su obrađeni QIIME 2 programom. Prema rezultatima ovog rada nije uočen statistički značajan utjecaj ITM i životnog stila na sastav i bogatstvo crijevne mikrobiote na promatranoj grupi ispitanika. Obesity, which has become prevalent in recent years, is associated with development of chronic diseases and increased mortality. Because of that it is important to analyse factors that have impact on obesity development. Some studies point to altered gut microbiota in obese subjects. The aim of this study was to analyse the impact of Body Mass Index (BMI) and lifestyle factors on gut microbiota. Subjects filled out a general questionnare containing lifestyle questions. Study included 15 underweight (BMI < 20 kg mˉ²) and 25 overweight (BMI > 27 kg mˉ²) subjects. Gut microbiota composition was determined by isolation of DNA from fecal samples which was then sequenced by Illumina MiSeq technology. Raw sequencing data were proccesed using QIIME 2. The results show no statistical significance of BMI and lifestyle factors on composition and richness of gut microbiota in test subjects.
