They are debilitating diseases and affect, mainly, the population of poor regions. Due to the limitations of the existing treatment, the search for new drugs become urgent. In this sense, the study of plant species leads as an alternative to obtaining a phyto-pharmaceutical for their treatment and control. Many Piper species are used for curative purposes by various cultures and they are composed by unsaturated amides, flavonoids, lignans, aristolactams, long/short-chain esters, terpenes, steroids, propenyl phenols and alkaloids. Piper marginatum is also known as capeba, malvarisco, wild pepper, capeba-fragrant and nhandi. It is popularly used against liver and vesicular disease, snakes and insects bites. Volatile compounds and Piper marginatum were investigated by determining the chemical constitution, seasonal variation and anti-protozoan in vitro activity. Essential oils were extracted by hydro-distillation, for 6 hours, and yields ranged from 0.10-0.23% v/m. Essential oil components have been identified through gas chromatography analysis coupled to a mass spectrometry (GC/MS), the main constituents were: ??-3-careno, (Z) ??-ocimene, ??-ocimene, ??-terpinolene, Ecaryophyllene, biciclogermacrene D, ??-copaene, linalool, myristicin, 3.4-(methylenodioxy) propiophenone. This is the first work that describes in vitro activity of the essential oil of P. marginatum against adult forms of S. mansoni, trypomastigote forms of T. cruzi and promastigotes forms of L. amazonensis. P. marginatum oil presented a promising schistosomiasis activity, reducing the motility and causing death of the parasites directly proportional to the concentration and incubation time. When tested at concentrations of 12.5 ??g/mL, this oil was capable to causes 100% of the motor activity of worms decreased after 24 hours of incubation and 100% death of the worms in a concentration of 50 ??g/mL after 120 h of incubation. For T. cruzi, the essential oil shown to be active in the concentration of 20 g/mL, resulting in 71% mortality of parasites, however the selectivity index was below 2.3, far below the selectivity index (less than 50) considered for in vivo tests. For the infectious forms of L. amazonensis, the oil proved to be inactive. The results obtained in this study against the S. mansoni adult worms, demonstrate the importance of phytochemical study of species found in the Amazon region, as an alternative to the existing restricted therapy to treat schistosomiasis. Muitas das esp??cies de Piper s??o usadas para fins curativos por diversas culturas, a sua constitui????o ?? composta por amidas insaturadas, flavonoides, lignanas, aristolactamas, ??steres de cadeias longas e curtas, terpenos, esteroides, propenilfenois e alcaloides. Piper marginatum ?? conhecida como capeba, malvarisco, pimenta-do-mato, capeba-cheirosa e nhandi. Ela ?? utilizada popularmente contra doen??as hep??ticas, vesiculares, picadas de cobras e insetos. Os compostos vol??teis de Piper marginatum foram investigados atrav??s da determina????o da constitui????o qu??mica, varia????o sazonal e atividade antiprotozo??ria in vitro. Os ??leos essenciais foram extra??dos pelo m??todo de hidrodestila????o, por seis horas, e os rendimentos variaram de 0,10 - 0,23% v/m. Os componentes do ??leo essencial foram identificados atrav??s da an??lise por cromatografia gasosa acoplada a espectroscopia de massas (CG-EM) e os principais constituintes foram: ??-3-careno, (Z)??-ocimeno, (E)??-ocimeno, ??-terpinoleno, Ecariofileno, germacreno D, biciclogermacreno, ??-copaeno, linalol, miristicina, 3,4- (metilenodioxi)propiofenona. Este ?? o primeiro trabalho que descreve atividade in vitro do ??leo essencial de P. marginatum contra formas adultas de S. mansoni, formas tripomastigotas de T. cruzi e formas promastigotas de L. amazonensis. O ??leo de P. marginatum apresentou uma promissora atividade esquistossomicida, reduzindo a motilidade e causando morte dos parasitas de forma diretamente proporcional ?? concentra????o e ao tempo de incuba????o. Quando testado na concentra????o de 12,5 ??g/mL, este ??leo foi capaz de causar 100% da diminui????o da atividade motora dos vermes ap??s 24 horas de incuba????o e morte de 100% dos vermes na concentra????o de 50 ??g/mL ap??s 120 h de incuba????o. Para T. cruzi, o ??leo essencial mostrou-se ativo na concentra????o de 20 ???g/mL obtendo um percentual de mortalidade de 71% dos parasitos, por??m o ??ndice de seletividade foi menor que 2,3, muito abaixo do ??ndice de seletividade (maior que 50) considerado para testes in vivo. Para as formas infectivas de L. amazonensis, o ??leo testado mostrou-se inativo. Os resultados obtidos neste estudo contra os vermes adultos de S. mansoni, evidenciam a import??ncia do estudo fitoqu??mico de esp??cies encontradas na regi??o Amaz??nica, como alternativa ?? restrita terap??utica existente para o tratamento da esquistossomose.
