For this, biotechnology provides some tools such as calli in vitro culture, to obtain substances of interest. Current studies with Deguelia duckeana revealed anticancer potential of flavonoids isolated from the roots of the species. In this work, we initially aimed to establish and in vitro culture of the plant species. Once callus obtained, they were dried and extracted with hexane and ethyl acetate. The extracts were fractionated to isolate the substances produced by the callus. As results obtained are that: in vitro callus establishment of this species was possible, aseptic cultures were obtained from young leaves of D. duckeana that underwent a disinfestation process, followed by induction and multiplication in MS medium added of 3 mg.L-1 of BAP + 2 mg.L-1 of NAA + mg.L-1 of 2 Kin. The calli were dried in lyophilizer and the 1: 1 combination of the hexane/ethyl acetate solvents proved to be the best for extracting the present metabolites. Fractionation of hexane/ethyl acetate 1:1 extract from callus allowed to isolate the triterpenes 3??-hydroxy-20(29)-lupen-28-al (betulinaldehyde) and lupeol. Their structures were confirmed by analysis of 1H and 13C NMR spectra, and COSY, HSQC and HMBC contour maps. Para isso a biotecnologia fornece algumas ferramentas como o cultivo de calos in vitro, com a finalidade de obter subst??ncias de interesse. Estudos atuais com Deguelia duckeana revelaram potencial antic??ncer de flavonoides isolados a partir das ra??zes da esp??cie. Neste trabalho, objetivamos inicialmente realizar o estabelecimento e o cultivo in vitro da esp??cie vegetal. Uma vez obtidos os calos, estes foram secos e extra??dos com hexano e acetato de etila. Os extratos foram fracionados, a fim de isolar as subst??ncias produzidas pelos calos. Como resultados obtidos est??o que: o estabelecimento de calos in vitro dessa esp??cie foi poss??vel, as culturas ass??pticas se deram a partir de folhas jovens de D. duckeana que passaram por um processo de desinfesta????o, seguido de indu????o e multiplica????o em meio MS adicionado de 3 mg.L-1 de 6-benzilaminopurina (BAP) + 2 mg.L-1 de ??cido 1-naftaleno ac??tico (ANA) + 2 mg.L-1 de cinetina (Kin). Os calos foram secos em liofilizador e a combina????o de 1:1 dos solventes hexano/acetato de etila se mostrou a melhor para extrair os metab??litos presentes. O fracionamento do extrato hexano/acetato de etila 1:1 dos calos permitiu isolar os triterpenos 3??-hidroxi-20(29)-lupen-28-al (betulinalde??do) e lupeol. Suas estruturas foram confirmadas atrav??s da an??lise dos espectros de RMN de 1H e de 13C, e mapas de contorno COSY, HSQC e HMBC. |
