Among the diseases that have an economic impact on guarana is oversprouting that affects plant growth and development, which is characterized by three different symptoms depending on the affected tissue: 1- stem galls, 2- oversprouting of vegetative buds and 3- oversprouting of inflorescences, these symptoms compromise productivity and can cause losses of up to 100%. In order to identify and characterize anatomical, histological and molecular alterations, comparative analyzes in symptomatic and asymptomatic tissues were carried out based on different approaches such as: light and electron microscopy, genomic, transcriptomic and proteomics. The morphoanatomical analyzes reveal that the different symptoms present a marked reduction in the longitudinal axis and the formation of ???capsules???, with thick and hard outer walls. When the infection affects the inflorescences, extremely malformed floral organs with multiple flowers within a single flower are also observed, and in the vegetative buds the vegetative primordia are poorly differentiated, with the formation of multiple malformed organs and an axillary meristem encapsulated by very thickened bracts. On the other hand, the aggregation of multiple capsules in the same branching point produces the symptom called gall. The morphological aspects observed in the symptoms suggest a possible hormonal imbalance, which was corroborated by the identification in the complete genome of the pathogen of genes for auxin production pathways (AIA) in F. decemcellulare and subsequent metabolomic analysis that confirmed the production. These first indications, together with the data of differentially expressed genes in the transcriptome comparing symptomatic and asymptomatic tissues of the three different symptoms of the disease, provided the first indications of the molecular mechanisms of action of the pathogen and the consequences of hormonal imbalance are presented in chapter I of this thesis. For a better understanding of the mechanisms that trigger the disease in guarana, the interaction proteome was also obtained, where 3404 guarana proteins were identified, of which 253 were differentially abundant between diseased and healthy tissue. These results confirm the transcriptomic observations and reinforce the role of the imbalance caused by auxin in oversprouting. All proteins of the SCF mechanism (Skp1???Cullin???F-box) of auxin response were identified and the differential abundance of some SCF components and proteins related to AIA conjugation in diseased tissue reinforce the role of imbalance in infected tissue. Difference in the abundance of proteins such as IAR3, IAGLU and AGO10 that allow greater ability in irreversible conjugation (AIA-Asp and AIA-Glu) and Argo10 related to "downstream" modulation of the auxin response with consequent restoration of hormonal balance are more effective in genotypes with higher level of resistance. O superbrotamento ?? uma doen??a causada pelo fungo Fusarium decemcellulare que atinge o guaranazeiro (Paullinia Cupana var. sorbilis), uma esp??cie nativa da Amaz??nia, utilizada amplamente pelas ind??strias de bebidas, cosm??tico e farmac??utica. Entre as doen??as que apresentam impacto econ??mico em guaranazeiro est?? o superbrotamento que afeta o crescimento e desenvolvimento da planta. Dependendo do tecido afetado, os principais sintomas: s??o galhas no caule, superbrotamento das gemas vegetativas e superbrotamento das infloresc??ncias. Estes sintomas comprometem a produtividade podendo causar perdas de at?? 100%. Com o objetivo de identificar e caracterizar as altera????es anat??micas, histol??gicas e moleculares, an??lises comparativas em tecidos sintom??ticos e assintom??ticos foram realizadas com base em diferentes abordagens como: microscopia de luz e eletr??nica, gen??mica, transcript??mica e prote??mica. As an??lises morfoanat??micas revelam que os diferentes sintomas apresentam uma redu????o acentuada no eixo longitudinal e a forma????o de ???c??psulas???, com paredes externas espessas e duras. Quando a infec????o afeta as infloresc??ncias tamb??m s??o observados m?? forma????o nos ??rg??os florais com m??ltiplas flores dentro de uma ??nica flor, baixa diferencia????o dos prim??rdios vegetativos nas gemas vegetativas e surgimento de m??ltiplos ??rg??os malformados e um meristema axilar encapsulado por br??cteas muito espessadas. J?? o agregado de m??ltiplas c??psulas em um mesmo ponto de ramifica????o produz o sintoma denominado de galha. Os aspectos morfol??gicos observados nos sintomas sugerem um poss??vel desequil??brio hormonal, o que foi corroborado com a an??lise gen??mica do pat??geno para vias de produ????o de auxina (AIA) em F. decemcellulare e sua posterior confirma????o por an??lises metabol??micas. Os dados comparativos da an??lise transcript??mica dos tr??s diferentes sintomas da doen??a s??o apresentados no cap??tulo I desta tese. Esses resultados forneceram os primeiros ind??cios dos mecanismos moleculares de a????o do pat??geno e as consequ??ncias do desequil??brio hormonal no guaranazeiro. Para melhor compreens??o dos mecanismos que desencadeiam a doen??a em guaranazeiro foi realizada uma an??lise prote??mica comparativa entre tecido doente e sadio. Um total de 3404 prote??nas de guaranazeiro foram identificadas, sendo 253 diferencialmente expressas entre as condi????es. Estes resultados confirmam as observa????es transcript??micas e refor??am o papel do desequil??brio causado pela auxina no superbrotamento. Todas as prote??nas do mecanismo SCF (Skp1???Cullin???F-box) de resposta a auxina foram identificadas e a abund??ncia diferencial de alguns componentes do SCF e de prote??nas relacionadas a conjuga????o de AIA no tecido doente refor??am o papel do desequil??brio no tecido infectado. A an??lise das prote??nas diferencialmente abundantes tamb??m sugere a perda da capacidade de regula????o da auxina nos tecidos infectados, bem como o aumento da express??o de prote??nas relacionadas ?? bioss??ntese do ??cido gama-aminobut??rico (GABA). E em outras esp??cies de plantas o ac??mulo de GABA desencadeia a forma????o de sintomas semelhantes ao superbrotamento. A an??lise de prote??nas de F. decemcellulare expressas durantes a intera????o pat??geno-hospedeiro identificou 31 prote??nas, entre elas 5 (cinco) correspondem a prote??nas efetoras, e uma a espermidina sintase que ?? uma enzima-chave na bioss??ntese de poliamina, uma mol??cula relacionada ?? s??ntese de GABA.
