Maternal obesity increases lipogenesis and nonalcoholic fatty liver disease in the offspring until the F2 generation

The present study evaluated the effects of maternal obesity upon liver structure in early adult life, focusing on the F1 and F2 generations. C57BL/6 female mice (F0) were fed standard chow (SC) or HF diet (8 weeks) prior to mating and during the gestation and lactation to provide the F1 generation (F1-SC and F1-HF). At 3 months old, F1 females were mated to produce the F2 generation (F2-SC and F2-HF). The liver was dissected, weighed and sliced into several fragments at 5 μm thickness and stained with hematoxylin and eosin. Frozen liver tissue was used to analyze western blot and triacylglycerol. Data were analyzed by unpaired Student s t-test. The HF-F1 group had higher body mass (BM) at 90 days when compared with SC-F1 (P=0.001), whereas the HF-F2 offspring showed no significant difference in BM when compared to SC-F2 offspring. HF-F1 had glucose intolerance when compared to SC-F1 group. This result was not observed in the F2 offspring. Nevertheless, HF-F1 offspring (P=0.009) and the HF-F2 showed hyperinsulinemia (P=0.03).
