The dissertation work examines the development of the proteinuria and the frequency of glomerular and tubular lesions as a function of the post-transplant time in a collective of 25 kidney-transplanted patients. The nephrotoxic influence of the two immunosuppressive agents, Ciclosporin A and FK 506, is defined and compared. The influence of donor age and time of ischaemia on the renal proteinuria is investigated. Possible application of the SDS polyacrylamide gel electrophoresis for long-term monitoring of kidney-transplanted patients is discussed. A comparison of the results of this method with those of the conventional parameters serum creatinin, creatinin clearance, and the total protein concentration in the urine clarifies the value of SDS PAGE in the diagnostics of renal lesions after kidney transplantation. The results of the study show that semi-automatic gradient gel electrophoresis with micro-gels and subsequent densitometric analysis is a reproducible and highly sensitive method for the monitoring of kidney-transplanted patients. Repeated application of the analysis can help detecting occurrence or increase of glomerular and/or tubular lesions and, if necessary, initiate an appropriate therapy. |
