This thesis is an investigation into the expression of chorion gonadotrophin (CG)-like molecules in early bovine conceptuses. Past work from this laboratory reported low level production of luteinizing hormone (LH)-like mRNA by early bovine conceptuses. Based on homology between published LH and CG sequences in both primates and equids, it was decided to determine if the LH-like mRNA molecules expressed by bovine conceptuses are also CG-like. Reverse transcription-polymerase chain reaction (RT-PCR) was carried out on RNA from early bovine conceptuses using both $\beta$LH and degenerate CG primers. The fragment amplified with $\beta$LH primers was sequenced and identified to be homologous with the published bovine LH sequence. Fragments amplified with degenerate CG primers were cloned into a plasmid vector and sequenced; there was no convincing overall homology with published LH or CG gene sequences. The fragment amplified with the degenerate CG primers exhibited considerable sequence homology to a region of glia-derived nexin genes (human 84%, rat 82%), expression of which has not previously been reported for bovine conceptuses. |
