Enzyme immunoassay for rat C-peptide

Popis: Our previous studies suggest that C-peptide augments glucose utilization in diabetic rats. However, a reliable assay for rat C-peptide has not been available. In the present study, a sensitive and specific enzyme immunoassay for rat C-peptide has been developed. Effects of rat, pig, goat and fetal bovine sera on the standard curve were studied, and did not produced significant displacement. Rat C-peptide standard concentration ranged from 0.024-100 ng/ml. The intraassy coefficient of variation (CV) was 3.9 ± 2.2% (less than 8%). The fasting and postprandial plasma C-peptide level in normal rat were 6.89 ng/ml and 13.28 ng/ml, respectively. Rat plasma C-peptide levels following oral glucose tolerance test in normal rats were consistent with expected patterns. In conclusion, we have developed and validated a rat C-peptide enzyme immunoassay that is sensitive and simple for rat C-peptide in plasma. This assay provides a reliable tool for studies of diabetes using rat models.
