EFFECT OF LINOLEIC ACID ON ANTI-TUMORAL EFFECT OF 5-FLUOROURACIL

Popis: Linoleic acid (LA) has been reported to have potential in selecting non-proliferative cancer stem cells. We studied the effects of linoleic acid on the anti-tumor effect of 5-fluorouracil (5-FU). Examining the 5-FU sensitivity in mouse lung cancer cell line LL2, and mouse colorectal cancer cell lines CT26 and CMT93, CMT93 showed low sensitivity. The increased growth inhibitory effects in CT26, CMT93, and LL2 were 2.5-, 1.4-, and 2.6-times, respectively, on concurrent treatment with LA at an apoptosis-inducing concentration and 5-FU at a concentration of IC50. On examining aldehyde dehydrogenase (ALDH) activity based on changes in the cancer stem cells in this case, it was found to have increased 1.7- to 3.2-fold in response to concurrent treatment with 5-FU and LA in the 3 cell lines. Further, the expression of nucleostemin, a stem cell marker, showed almost no change between simultaneous treatment with 5-FU and LA, and treatment with LA alone. When used in conjunction, LA appears to be enhancing the 5-FU Sensitivity of cancer cells; however, it may increase the survival of cancer stem cells. These results suggest that the content of the diet during chemotherapy is critical in optimizing the host’s condition.
