A zinc finger protein, Zfp444, is expressed in differentiating pancreatic β-cells and activates Glut2 gene expression

Popis: It is generally believed that the pancreatic β -cells are derived from ductal epithelial cells during fetal and neonatal life. It is critical to elucidate its mechanism for the regeneration medicine of diabetes. Here, using laser capture microdissection and differential display methods, we isolated cDNAs that are expressed predominantly in putative differentiating β -cells which are positive for both cytokeratin 19 (a marker of ductal cells) and insulin in a higher degree than ductal cells. Among those RNAs, we identifi ed a zinc fi nger protein, Zfp444, which is expressed more in the pancreatic β -cell lines than in a ductal cell line, and more in fetal pancreas than in adult pancreas. In situ hybridization and immunohistochemistry revealed that it locates in the pancreatic islet cells expressing both insulin and cytokeratin 19 in the fetal pancreas. Furthermore, Zfp444 increases the Glut 2 mRNA level. Taken together, these results suggest its possible role in β -cell generation.
