The influence of cationic, anionic, amphiphilic and neutral membrane-active agents on electrofusion of barley protoplasts in a meander chamber - after dielectrophoresis - has been studied. From the fusion yield dependence on concentration of these agents the following types of curve have been obtained: 1. by anions: decreasing yields as concave curves; 2. by cations: decreasing yields at higher concentrations as convex curves, owing to the dissipation of pulse energy by increasing conductivity, 3. by amphiphilic agents: increasing curves for fusion yield at low concentration, 4. by neutral agents: exponentially increasing fusion yield depending on adsorptivity and molecular mass. In spite of the low polymer concentration ( ⪡ 4%) applied, fusion yields can be increased by 90%. In line with this trend of synergistic effects between membrane active agents and electrofusion itself, the pulse field strength and width can be lowered remarkably in order to minimize side-effects on cell metabolism and viability. |
