Popis: | Serum thyroxine (T4) levels were measured in serum pools by competitive protein binding radioassay (CPBR) with four kit procedures and an independent method. Thyroxine values in the pools analyzed were stable for three days at 25° and for l h at 45°. Thyroxine values obtained with the Abbott, Curtis and Mallinckrodt methods were greatly affected by the length of the separation step incubation, while values obtained with the Nuclear Medical method were least affected by the length of this step. Only with the Abbott procedure were coefficients of variation below 10% obtained for T4 values at all T4 concentrations employed. Recoveries of added T4 were high with the Abbott method and low with the Curtis method. Recoveries were between 80 and 100% with the Mallinckrodt method. Dilution of the serum pools we used with saline resulted in an increased extraction efficiency of T4. |
