Popis: | Possible tissue differences in the content of aminoacyl-tRNA synthetases specific for degenerate tRNA species for the same amino acid were investigated in the rabbit through measurements of PP i -dependent deacylation of aminoacyl-tRNA. Using a double-label procedure, comparisons were made of the relative activity of a given enzyme preparation towards a tRNA mixture previously acylated either by that same preparation (homologous deacylation) or by a different preparation (heterologous deacylation). Marked differences in homologous vs. heterologous deacylation could be readily demonstrated with two partially purified leucyl-tRNA synthetase preparations known to differ in specificity in the forward reaction, however, little evidence was found for differences in tissue synthetase specificity among crude, unfractionated preparations from four tissues. The synthetases for seven amino acids were surveyed. The evidence suggests that presence-absence differences between tissues in codon-specific aminoacyl-tRNA synthetases are rare or generally confined to tRNA species or synthetases present in low concentrations, undetected by the present method. |
