Popis: | Summary The decay of mRNA's corresponding to the argECBH gene cluster of Escherichia coli has been studied by RNA-DNA hybridization methods. In the argA−argR+ strain 39A-23R3, the mRNA's were labeled with tritiated uridine either during message accumulation by arginine starvation or during growth under conditions derepressive for the arginine system. The decay of the messages was followed, in the presence of a repressive ( l -arginine hydrochloride) or a restrictive (α- N -acetyl- l -ornithine) arginine source, by measuring the disappearance of radioactive label after a chase with cold uridine or after treatment of the cells with rifampicin or after both. Accumulated argECBH messages show half-lives of 1.5 and 4.3 min for arginine excess and arginine restriction, respectively. Under conditions of growth, i. e., under physiological conditions, the corresponding half-lives are 1.6 and 6.6 min. |
