Non-steroid inhibitors of pituitary gonadotrophic function

Popis: Nonsteroid inhibitors of pituitary gonadotropic function were studied. The most active of the compounds examined was 1-alpha-methylal lyltheocarbamoyl-2-methyl-thiocarbamoylhydrazine. This substance caused selective and reversible inhibition of pituitary gonadotropic function in male and female rats. Similar evidence was obtained in dogs and monkeys but not in mice guinea pigs rabbits or horses. The effect of this compound on the reproductive system of rats was apparent with daily doses as low as 16 mg/kg but daily doses of 350 mg/kg were tolerated for up to 6 months without serious effects on well being or on the morphology of other internal organs. It is concluded that the compound either directly interferes with the production of gonadotropins by the pituitary or affects the presumed hypothalamic control of that function.
