Prostatic tissue distribution of 17β-hydroxy-5α-androstan-3-one and of androgen receptors in benign hyperplasia

Popis: | Stromal-epithelial relationships may be involved in the androgen-dependent development and maintenance of benign hyperplasia in the human prostate. Concentrations of endogenous 17β-hydroxy-5α-androstan-3-one (DHT) have been determined in separated epithelial and stromal components of hyperplastic prostate. Androgen receptor concentrations have also been measured in cytosols and nuclear extracts prepared from both tissue components, using conditions which allow detection of occupied receptor sites. The results indicate that cytosolic and nuclear androgen receptors are found in both epithelium and stroma, and that there is parallel distribution of 17β-hydroxy-5α-androstan-3-one. There is no substantial enrichment of receptor in epithelial cells compared to the stromal component. Both stroma and epithelium possess an androgen receptor capable of entering the nucleus and are therefore potentially androgen sensitive. |
