Mechanisms of macrophage activation by Corynebacterium parvum

Popis: Normal CBA mouse peritoneal macrophages could not be activated, as assayed by their subsequent inhibition of syngeneic leukemia cell DNA synthesis, by exposure to Corynebacterium parvum , but only by mixtures of C. parvum and spleen cells from C. parvum -immune mice. Treatment of immune spleen cells by anti- θ serum and complement abolished activation. Supernatants from cultures of immune spleen cells and C. parvum activated normal macrophages, and the effect was potentiated by adding C. parvum or other bacteria during the treatment of the macrophage with supernatants. A strain of C. parvum and of C. diphtheriae , lacking RE-stimulating activity and adjuvanticity in vivo , were less effective both as producers of immune spleen cells and in cooperating with C. parvum -immune spleen cells in generating activation in vitro . Oil-induced macrophages were activated by C. parvum alone, and by the coryneforms inactive in vivo . It is concluded that C. parvum may activate macrophages in vitro by both direct and immunological mechanisms.
