Popis: | The rates of synthesis and degradation of catalase protein were measured in cultures of human diploid fibroblasts by the method of Price et al. [10] which employs 3-aminotriazole as an irreversible inhibitor of catalase without affecting its synthesis. The half-life of catalase has been estimated at approx. 30 h, a value comparable to that obtained for rat and mouse liver catalase. The turnover of catalase is influenced by a factor present in fetal calf serum which is inactivated during incubation. Suboptimal amounts of this serum factor retard both synthesis and degradation of catalase, resulting in a reduction in the cell content of catalase. From the double isotope method of Arias et al. [18] it would appear that catalase is turning over more slowly than average cellular protein. |
