The method of neutrophils activity description

Popis: In some immunodeficiency disorders ineffectiveness of neutrophils' motility is affected. As the background of new diagnostics method the new method of description of single cell's activity is presented. This method analyses three aspects of its behaviour: penetration intensity, directionality and expansiveness of movement. The proposed method of cell's activity and cells motility evaluations is adjusted to the Cells Movement Monitoring System, in which neutrophils' behaviour is documented as a sequence of bright-field microscopic images. The cell's positions on the plane and static area of its activity are examined in each cell's static position. These static parameters are estimated based on cells outline. Based on variation in time of these static parameters several dynamic properties are calculated. The definitions of the cell's positional centre of gravity in the lamina and the wire models of the cell and the static cell activities area of the elliptical model of the cell lead to the smallest variation of activity description and motility evaluation. The method of cell's behaviour description and cells' population motility evaluation has been tested on selected sequences from experimental material. The behavioural description agrees with the subjective visual human verification.
