Popis: | Rat yolk sacs were obtaine about 18 days post coitum . Incubations invitro were carried out and the following observations were made: (I) Incorporation of labeled glycine and glucose into protein and nucleic acids was markedly inhibited by the antibiotics puromycin and d - threo -chloramphenicol. The antibiotically inactive isomers of chloramphenicol also exerted inhibitory activity, but ot varying degrees. (2) Puromycin has no effect on O 2 uptake. Chloramphenicol exerted marked inhibitions of O 2 uptake, which seems to account for the degree of inhibition of protein and nucleic acid synthesis. (3) The sensitivity of yolk sac to d - threo -chloramphenicol was not found in the topologically similar tissue, rat diaphragm. (4) Apparent entry of 14 C-labeled homologous hemoglobin into yolk sac was studied in vitro . Anaerobiosis, 2,4-sinitrophenol, chloramphenicol, puromycin, and incubation in the cold all caused significant inhibitions. The evidence indicated that at least part of the protein entry was dependent on supplies of metabolic energy. |
