Popis: | The use of the metal-ion indicator, Arsenazo III, for the spectrophotemetric measurement of total and free calcium concentrations has been explored. The indicator can be used satisfactorily in the physiological pH range. A procedure is described for the direct measurement of calcium-binding equilibria in a ternary system containing the indicator, calcium ions, and a calcium-binding macromolecule. This appears to offer some advantages over conventional methods in situations in which the total calcium-binding capacity is low. Results on two proteins are reported, and the limitations of the method are discussed. |
