Popis: | A simple procedure is described for using commercially available recording spectrophotometers for determining absorption spectra of aqueous samples at low temperature. In contrast to previous methods, very rapid initial freezing is employed followed by scanning of the specimen in a windowless metal cuvette maintained near the boiling point of liquid N2 in an inexpensive, easily constructed, polyurethane foam chamber. Sharpening, splitting, and intensification of absorption bands of cytochrome c and benzene in water are described. The windowless cuvette has been used for the 220–800-mμ wavelength range. |
