Immunoglobulin classes, IgG subclasses, Gm genetic markers, and Clq following bone marrow transplantation in X-linked combined immunodeficiency

Popis: | Patient D. C. with X-linked lymphopenic immunodeficiency (LID) underwent successful immunologic reconstitution by bone marrow transplantation at age 5 months. Quantitations of Ig classes, IgG subclasses, Gm genetic heavy chain markers, and Clq component of complement during the subsequent 4 1/2 years were compared with data of the donor, parents, and age-matched controls. A striking rise in all five classes of immunoglobulins in D.C. was noted. IgG1 and IgG3 subclasses rose significantly in excess of normal without evidence of monoclonal synthesis. IgG2 levels prior to transplantation were normal and exhibited a lesser rise. The IgG1 and IgG3 genetic markers Gm(f) and Gm(b) rose in parallel with their respective subclasses and the IgG2 marker Gm(n) rose more slowly. Within three months after transplant the Gm markers Gm(a) and Gm(g) disappeared, reflecting either catabolism of administered gamma globulin or maternal gene complex markers absent in the donor. The synthesized Gm markers Gm(f+b+n+) corresponded to those of the donor. the Clq component of complement exhibited a rise and subsequent return to normal levels paralleling and slightly antedating the changes observed in concentrations of immunoglobulins known to bind to Clq. |
