Toxoplasma: Seroreactions with antibody fragments

Popis: Toxoplasma (TP) Seroreactions were studied with antibody fragments derived from rabbit anti-TP γG-immunoglobulin. To assess the role of bivalence and the significance of the Fc fragment in TP Seroreactions, 5S bivalent antibody fragment [F(ab′) 2 ]was prepared by the pepsin digestion of γG-immunoglobulin, and 3.5S monovalent antibody fragment (Fab′) was prepared by the reduction and successive alkylation of the 5S fragment. The fragments and original γG-immunoglobulin were tested for activity in the hemagglutination test (HAT), dye test (DT), and complement fixation test (CFT). The γG-imnunoglobulin elicited positive reactions in every one of these tests. The 5S fragment elicited positive reactions in HAT and negative reactions in CFT and DT. The 3.5S fragment failed to elicit positive reactions in any of these tests, although the potential of the 3.5S fragment to react with TP antigen was demonstrated by Coombs' test and by absorption with TP cells. The results indicate the important role of bivalent antigen-binding in HAT and the indispensable role of the Fc fragment in CFT and DT. The significance of the Fc fragment in DT is discussed in reference to the complement-like function of accessory factor.
