A new method for the continuous selection of large numbers of cells in mitosis from a monolayer culture grown in a rotating bottle is described. It has so far been applied to HeLa cells. The mitotic cells are brought into suspension by a smooth, mechanical shaking, and are continuously trapped at a subnormal temperature. By varying the shaking frequency, the circulation velocity and the glass surface, it is possible to harvest the cells in different phases of mitosis. When transferred to a temperature of 37 °C again, the cells are able to proliferate. The relatively low degree of synchrony which appears in the process must be partly due to the great heterogeneity of the HeLa cell culture. |
