Initial rates at pH 6.5 and 23 ° of chymotrypsin-catalyzed condensations of N-benzoyl- l -tyrosine (NBT) with glycylanilide (GA) to yield N-benzoyl- l -tyrosylglycylanilide (NBTGA) show marked induction periods despite the insolubility of NBTGA. Scratching to provide crystallization nuclei shortens the induction period, and inclusion of an NBTGA suspension in the reaction system eliminates the induction period. The effect of initial concentrations of reactants on initial rates of synthesis was determined by following disappearance of the amine moiety, NBTGA being included in the reaction system. Inital rates show a Michaelis-Menten dependency on NBT concentration with KNbt 0.08 M and Vmax 1.5 × 103 mole/l./mole enzyme/mole GA. Initial rates are linearly dependent on GA concentration. These results are interpreted as indicating the synthetic process is the reverse of the accepted scheme for chymotrypsin-catalyzed hydrolysis. |
