Popis: | Trihexosylceramide biosynthesized in vitro from lactosylceramide and UDP-galactose in the presence of a galactosyltransferase from rat spleen microsomes or bone marrow was shown by methylation to have the structure galactopyranosyl-α-(1 → 3)-galactopyranosyl-β-(1 → 4)-glucopyranosyl-β-(1 → 1)-ceramide. It is thus different from trihexosylceramide isolated from several natural sources in which the two galactoses are linked 1 → 4. This emphasizes the need for establishing by unequivocal means the structure of glycolipids biosynthesized in vitro. The method described permits this to be done on the very small amounts of material usually available from biosynthetic experiments. |
