A calorimetric investigation of tRNA1Val melting

Popis: Melting of purified tRNA1Val was investigated by scanning microcalorimetry under various salt conditions. It is shown that the total enthalpy of melting is 370 kcal mol−1 and depends only slightly on salt conditions and temperature. Such a high enthalpy is in good agreement with recent results of structural analysis, according to which more than 82% of bases are bonded in native tRNA. The thermodynamic analysis of the complicated melting curves shows that there are six essentially independent co-operative regions in tRNA structure; some of these regions merge in the presence of magnesium ions. A structural description of these regions is given.
