Carbon isotope(s) enrichment of haemoglobin by specific biosynthetic labeling with amino acids

Popis: 1. 1. Reticulocytes, bone marrow cells and foetal liver cells, mainly from rabbit, were tested for incorporation of certain amino acids into haemoglobin molecule, using 14C-labeled amino acids. 2. 2. Best results were obtained with 18-day old foetal liver cells, and the efficiency of the older cells was markedly smaller. 3. Histidine was incorporated into up to 23% of the available sites, tyrosine to about 10% and aspartic acid to 5%. 4. The results indicate the feasibility of preparing haemoglobin sufficiently enriched also with 13C-labeled amino acids for nuclear magnetic resonance studies.
