Disposition of diethylstilbestrol in the rhesus monkey

Popis: The biliary excretion and disposition of diethylstilbestrol-14C was studied in an adult male rhesus monkey surgically prepared for chronic use with indwelling arterial-venous catheters as well as permanent catheters to the common bile duct and duodenum. Following intravenous administration, 23–37% of the dose was excreted into the bile after conversion to diethylstilbestrol monoglucuronide. When bile containing this metabolite was instilled into the duodenum, it was absorbed (to the extent of about 36%), following hydrolysis of the glucuronide, setting up the potential for enterohepatic recycling. Analysis of diethylstilbestrol blood levels indicated that this drug was rapidly metabolized and excreted following intravenous administration, having a total body clearance of 381 ml/min. Consistent with such a rapid clearance, the availability of diethylstilbestrol was very low following a duodenal dose in solution. A two-compartmental model best described the disposition of diethylstilbestrol characterized by the rapid clearance discussed above and a very large volume of distribution\((V_{d_{ss} } = 5 liters/kg)\).
