Popis: | The distribution of tolbutamide and chlorpropamide after i.v. administration in rats has been investigated. Tolbutamide or chlorpropamide did not accumulate in any of the organs studied, and autoradiographic investigations did not indicate any specific affinity to beta-cells. The highest concentrations, which at no time exceeded the concentration in the plasma, were found in liver and kidney. It would seem that tolbutamide penetrated the liver cells, but it was not possible to demonstrate cellular penetration in any other tissue. The distribution of chlorpropamide resembled that of tolbutamide, but uptake into the liver cells could not be demonstrated. Insulin altered the distribution of tolbutamide while it did not affect chlorpropamide in the concentration employed. |
